Hill Equation: Cooperative Oxygen / Receptor Binding & Hill Coefficient (nH)
Calculate fractional receptor or hemoglobin saturation θ (%) = 100 / (1 + (EC_50 / [L])^n_H) and arterial-to-venous cooperative O_2 unloading across positive (n_H > 1), non-cooperative (n_H = 1), and negative (n_H < 1) allostery.
How n_H = 2.8 allosteric cooperativity doubles hemoglobin’s tissue oxygen delivery!
Archibald Vivian Hill (Journal of Physiology, 1910) formulated the sigmoidal binding equation θ = [L]^n_H / (K_0.5^n_H + [L]^n_H) = 1 / (1 + (K_0.5 / [L])^n_H) to explain why tetrameric hemoglobin binds oxygen along an S-curve rather than a simple Michaelis-Menten hyperbola! When n_H = 1, binding sites act independently; when n_H > 1 (positive cooperativity, n_H = 2.8 for human adult HbA with P_50 = 26.6 mmHg), the first O_2 bound triggers the T-to-R conformational switch so remaining subunits snap up O_2 eagerly in the lungs—and dump it steeply in hypoxic muscle!
Quantify that physiological advantage between lung alveoli (PO_2 = 100 mmHg) and resting venous tissue (PO_2 = 40 mmHg) at P_50 = 26.6 mmHg: cooperative HbA (n_H = 2.8) loads to 97.61% in the lungs and drops to 75.81% in veins (unloading 21.80% capacity; plunging to 31.06% at exercising PO_2 = 20 mmHg!), whereas a non-cooperative n_H = 1 carrier with the same P_50 = 26.6 mmHg would load only 78.99% and drop to 60.06% (unloading just 18.93%)!
| Ligand / Oxygen Tension [L] (P50 = 26.6 mmHg) | Hill Coefficient nH | Fractional Saturation θ (%) | Allosteric Regime |
|---|---|---|---|
| Alveolar Arterial Blood (PO2 = 100 mmHg) | nH = 2.8 (Cooperative) | 97.61% | Near-Complete R-State Loading |
| Resting Venous Tissue (PO2 = 40 mmHg) | nH = 2.8 (Cooperative) | 75.81% (-21.80% Unload) | Steep Sigmoidal Shoulder |
| Hypothetical Non-Cooperative Site (PO2 = 40 mmHg) | nH = 1.0 (Hyperbolic) | 60.06% | Independent Langmuir/Michaelis |
Hill Fractional Receptor / Hemoglobin Saturation θ (%)
Evaluates 100 / (1 + (EC_50 / [L])^n_H) (returning 0% cleanly at [L] = 0 for positive n_H).

Computes the classic Hill-plot ordinate log_10(θ / (1 - θ)), whose slope vs log_10([L]) equals the Hill coefficient n_H.
